This is a mainly off road route, on the South Downs Way, that has a variety of surfaces and at least 3 major climbs. It is the reverse of the Eastbourne to Lewes (off Road) route previously posted, which has more detail about the varying types of terrain on the SDW.
I normally get the train to Lewes and ride back to Eastbourne, getting the advantage of the wind behind me. It's about 4 miles of A roads from Lewes to the South Downs Way, but once on this, it's a great off road ride. Just follow the official SDW signs. This ride should take just over 3 hours.
